Background:
Mesenchymal stem cells (MSC) are widely used in various clinical and medical experiments. Although there are high expectations for the efficacy of treating major diseases through stem cells, there is no guarantee of its safety. We conducted systematic clinical trials to comprehensively evaluate the safety of MSC.
Methods and results:
We looked at MEDLINE, EMBASE, and CENTRAL (through June 2011). We conducted clinical trials of vascular delivery (intravenously or intravenously) of MSC in adults and a mixture of adults and children. Studies that differentiated MSC and other cell types were excluded. Our main evaluation items included acute side effects (acute injection toxicity, fever), visceral complications, infections, and long-term adverse events (death and malignancy). We looked at 2,347 papers and examined 36 studies. The 1,012 participants included patients treated for ischemic stroke, Crohn's disease, myocarditis, myocardial infarction, graft-versus-host disease, and healthy individuals. A total of eight studies were randomized comparison trials (RCTS) involving 321 participants. In RCTS, we found no association between acute injection toxicity, organogenic comorbidity, infection, death, or malignancy. MSC was significantly associated with transient fever.
Conclusion:
At this stage of clinical trials, MSC therapy can be described as safe. However, to further ensure the safety of MSC, we will strictly report adverse events and conduct larger clinical trials.
